The FlowCytomix Multiplex Human Th1/Th2 10Plex kit is a multiplex fluorescent bead immunoassay for quantitative detection of the human cytokines; IFN-γ, IL-1β, IL-2, IL-4, IL-5, IL-6, IL-8, IL-10, TNF-α and TNF-β. Th1/Th2 cytokines are secreted by peripheral blood T and B lymphocytes, natural killer cells and monocytes as an antigenic response toward humoral or cell-mediated immunity. Automation of this multiplexed flow cytometry-based assay can increase the sample throughput and simplify the sample preparation process. The methods for this assay have been developed for the Biomek FX workstation with dual bridge, integrated Orbital Shaker ALP and SPE filtration unit.
The multiplex immunoassay kit includes a 96-well filter plate for efficient antibody staining and washing using the on-deck SPE filtration system. The detection and quantitation of multiple analytes are performed using the automated Cytomics FC500 MPL from Beckman Coulter for data acquisition and the FlowCytomix Pro software for data analysis. The automated FlowCytomix Multiplex Human Th1/Th2 10plex Assay provides a high-throughput, in vitro immunotoxicity assay for pre-clinical evaluation of candidate drug toxicity.
