SAGIAN Automated Assay Optimization, a new solution from Beckman
Coulter, breaks the bottleneck in assay development. Developed
exclusively for the Biomek® 2000 Laboratory Automation Workstation, AAO
is a powerful tool for rapidly developing robust, cost-effective
bioassays and transitioning assays from the bench to HTS formats.
SAGIAN AAO is advanced, incorporating proven statistical experimental
design, while at the same time practical, eliminating the mundane and
tedious task of manual pipetting or writing complex liquid handling
mehods.
With SAGIAN AAO, researches can harness the power of statistically
designed experiments to develop robust, cost-effective bioassays in days
rather than months.
Statistically Designed Experiments
Choosing from several assay templates, or creating your own, you can
then identify the inter- and intraplate factors to be studied such as
buffer composition or incubation time. One of four resolutions of
fractional factorial design is available for selection, as well as
experimental protocols such as quality control wells and well
restrictions. The result -- a targeted experimental design set for the
optimization study at hand.
Automatic Plate Mapping and Pipetting Methods
Once you design the experiment set, AAO automatically maps it into
randomized plate wells and facilitates creation of the Biomek 2000
methods. This includes the creation of buffer either in intermediate
labware or directly into the assay plates. You have complete control
over all pipetting parameters and the order of additions. After
inputting a few simple parameters, reagent liquid handling methods are
created seamlessly in the background.
Post-Assay Data Collection and Analysis
Once the assay has been completed data collection can be automated from
one of several plate readers or data can be imported into an Excel
spreadsheet. AAO automatically performs standard analyses to estimate
the effects of the various factors and interactions between factors. You
will be able to quickly see which factors are critical to assay
performance. You can then use this information for further in-depth
studies or to configure a robust, cost-effective assay.
The Results
Using AAO can shorten assay development time from months to weeks. At
the same time the quality of the assay improves -- generating better
leads.
